Dear all,
Following the E-mail of Cyril, it is indeed possible to measure the surface area of cells using ImageJ.
In order to do this you just need to download the following plugin: http://rsb.info.nih.gov/ij/plugins/afm.html
This plugin able you to import files from the Nanoscope Software (I extended this while I was working by Veeco and it should work for all the versions of the Nanoscope software) or from the Agilent Technologies facilities (previously Molecular Imaging).
Then you take the topography picture and make a threshold on the height in order to segment the different cells.
>From the obtained binary picture you just will need to measure the number of active pixels within your picture or ROI (Region Of Interest) and multiply it by the surface of a pixel and there you go.
