Hello,

With (free) software as Imagej, you can mesure aera pourcentage of 
occupation by the cells. With the total aera you obtain with gwiddion 
and the number of cells (if there are more than one) you can have the 
coverage average of cells.
This is a way inspired by biologist use, I hope it could help you.
