[Spm] How to calculate the surface area of the cell membrane?

Dear all,

I want to know how to calculate the surface area form the AFM topographic
image. As we all know, if we want to get the whole surface area we can use
the Gwyddion's statitistical function(by pressing the statistical quantities
button). My problem is: now I want to get the surface area of many specific
zones (eg: lipid raft) in the cell membrane respectively .

So, could anyone tell me what algorithm is used by Gwyddion to calculate the
surface area (or just describe the thought of the algorithm)?

Any suggesttion will be appreciated!*
