Dear Valeria,
Back in 1997, we wanted to perform indentation testing on cartilage
using larger contact areas. Therefore, we ordered diamond-like carbon
(DLC)-coated glass from Zeiss. The idea was to produce blunt AFM tips by
scratching at high force over the DLC surface. I remember scratching
half a day with a single Si-tip on the DLC coating, but the tip did not
get significantly blunt.
Next, we glued borosilicate glass spheres onto tipless cantilevers. If
you have a good micromanipulation stage (we are using Veeco’s Bioscope
AFM) you can glue spheres of down to about 1 micron diameter. Another
advantage is that you’re having defined tip geometry for calculation the
stiffness. Meanwhile, you’ll find several protocols for gluing spheres
on the internet. Also companies are now selling such spherical indenters…
I just thought while reading your email that you probably not want to
repeat my mistakes.
